Category: GLP-1/GIP/Glucagon Tri-Agonist | Half-life: 6 days

Mechanism

Triple incretin agonist. GLP-1 + GIP + glucagon receptor activation. Superior weight loss to Semaglutide/Tirzepatide. Phase 3 trials.

Dosing Protocol

Critical: Slow titration mandatory. Severe nausea/vomiting if escalated too fast. Not FDA-approved yet. Research only.

RETA-6X / RETA-12X | GLP-1 / GIP / Glucagon Triple Agonist | CAS: 2381089-83-2

Retatrutide is a next-generation triple incretin receptor agonist that simultaneously targets GLP-1, GIP, and glucagon receptors. It is designed to support significant fat loss through appetite suppression, increased energy expenditure, and improved insulin sensitivity. Clinical studies have demonstrated body weight reductions of up to 24%, making it one of the most promising compounds in metabolic health and weight management.

Dosing Protocol
ParameterDetails
Starting Dose2 mg subcutaneous injection once weekly
TitrationIncrease by 2 mg every 4 weeks as tolerated
Maintenance Dose6–12 mg per week (6X = 6 mg vial, 12X = 12 mg vial)
Administration RouteSubcutaneous injection in the abdomen, thigh, or upper arm
Cycle Length12–24 weeks, with reassessment after 12 weeks
ReconstitutionMix with 1–2 mL bacteriostatic water per vial
Stacking Recommendations

For enhanced growth hormone support, Retatrutide may be combined with Tesamorelin or a CJC-1295 and Ipamorelin protocol. For additional metabolic support, 5-Amino-1MQ may be considered. Concurrent use with other GLP-1 receptor agonists is generally not recommended. Supporting supplements such as Omega-3 fatty acids and Berberine may help maintain cardiovascular and metabolic health.

Note: Nausea is the most commonly reported side effect. Gradual dose escalation is recommended to improve tolerability. Regular monitoring of fasting glucose and lipase levels is advised. Avoid use in individuals with a history of pancreatitis.
